Luận văn Khảo sát ảnh hưởng của vi khuẩn Methylobacterium Sp. lên sự phát sinh cơ quan ở cây lúa (Oryza sativa L) nuôi cấy In Vitro

TÓM TẮT “ KHẢO SÁT ẢNH HưỞNG CỦA VI KHUẨN METHYLOBACTERIUM SP. LÊN SỰ PHÁT SINH CƠ QUAN CÂY LÚA (Oryza sativa L.) NUÔI CẤY IN VITRO”. Đề tài được thực hiện tại trại thực nghiệm – khoa Sinh học - trường Đại học Khoa học Tự nhiên trên đối tượng là giống lúa VĐ20 và chủng vi khuẩn Methylobacterium sp. 1019. Tiến hành nhân sẹo, tạo chồi, tạo rễ từ mô sẹo của giống lúa VĐ20 bằng phương pháp nuôi cấy in vitro, sau đó khảo sát ảnh hưởng của chủng 1019 lên khả năng phát sinh cơ quan của mô sẹo bằng cách bổ sung những nồng độ khuẩn khác nhau vào môi trường nuôi cấy với 34 nghiệm thức, mỗi nghiệm thức 3 lần lặp lại với các chỉ tiêu như tỷ lệ tái sinh, số chồi trên mẫu, số rễ trên mẫu, đường kính mô sẹo. Những kết quả thu được: - Chủng 1019 có tác dụng kích thích sự phát triển rễ, ức chế khả năng tái sinh chồi và thay đổi trạng thái, cấu trúc của mô sẹo. Chứng tỏ chủng khuẩn có tác động đến quá trình trao đổi chất, sinh lý, sinh hóa của tế bào mô sẹo, ảnh hưởng đến sự phát sinh hình thái của sẹo. - Chủng 1019 có ảnh hưởng đến quá trình phát sinh hình thái của giống lúa VĐ20, chiều hướng phát sinh cơ quan tuỳ thuộc vào bản chất của mô cấy và loài thực vật. - Nồng độ khuẩn cao sẽ ức chế hoàn toàn mô sẹo, không thấy được sự phát sinh cơ quan từ mô sẹo.
