Luận văn Đặc tính sinh lí, sinh hóa và một số công đoạn chính trong qui trình chế biến bột nhàu
Nguyên lý:
Định lượng vitamin C dựa vào tính khử của nó với thuốc thử 2,6-
diclorophenol indophenol. Dạng oxy hóa của thuốc thử này có màu xanh bị khử
bởi acid ascorbic có trong dung dịch chiết của nguyên liệu thực vật thành dung
dịch không màu. Ở điểm cân bằng tất cả acid ascorbic thì thuốc thử màu dư thừa
không bị khử có màu hồng trong dung dịch acid.
Tiến hành:
Cân 5g dịch nhàu sau khi xay chuyển sang cốc cùng với 20ml HCl 1%, cho
vào bình địng mức 100ml. Rửa cốc và tráng dụng cụ vài lần, mỗi lần với acid
oxalic 1% và cũng dồn vào bình định mức. Dùng acid oxalic để đưa thể tích lên
vạch 100ml. Lắc kỹ, để yên 15 phút rồi lọc qua giấy lọc khô.
Chuẩn mẫu đối chứng: lấy 8ml acid oxalic 1% 2ml HCl 1% cho vào bình
tam giác 100ml dùng microburet với 2,6-diclorophenol indophenol để chuẩn độ
đến lúc xuất hiện màu phớt hồng bền trong 1 phút.
